Hyperuricemia has known to be a risk factor for coronary artery disease, hypertension, and heart failure. Among angiotensin II receptor blockers(ARBs), only losartan lowers serum uric acid(UA) level compared with other ARBs. Recent studies have reported that UAcauses vascular smooth muscle cells(VSMCs) proliferation and inflammation by entering cells via a functional uric acid transporter(UAT). We investigated whether losartan could inhibit the UA-stimulated proliferative and inflammatory signaling activation by modulation of UAT in cultured rat aortic VSMCs. Following 72 hours incubation of UA, proliferation of VSMCs was increased dose-dependently. Losartan inhibited UA-stimulatedproliferation in dose-dependent manner with an IC50 value of about 1 M. Losartan attenuatedp38, ERKs activation, COX-2 expression, and MCP-1 production. Losartan significantly decreased expression of UAT in UA-stimulated VSMCs(33% reduction, p<0.05), which suggests inhibitory effect of losartan on the uptake of UA via UAT. These data provide that losartan may involve anti-inflammatory and antiproliferative activity by modulating UAT. Therefore, losartan may provide beneficial effect on vascular changes in hyperuricemic condition.

Schwann cell(SCs) has been widely accepted to play indispensable roles during neural development and regeneration but obtaining and culture of SCs is difficulty. SCs have been characterized depending on sequential passages and medium. We measured proliferation and phenotypical stability of SCs using three medium(growth medium, purification medium, high density culture medium). Morphological changes were observed by an optical microscope and cell proliferation was counted using hemacytometer. The effect of sequential passage and medium on the phenotypical stability of SCs was assessed RT-PCR and staining for NF and S-100 as SCs markers. In result, proliferation of SCs showed no difference between three groups up to passage 3. NF and S-100 markers were high expressed in up to passage 3 in PM and HDCM. We concluded that SCs of passage 1, 2 and 3 cultured in HDCM will be useful to cell therapy using SCs or spinal cord regeneration using SCs hybrid scaffold.

Porcine small intestine submucosa is accellular tissue and widely used as a biomaterials without immunorejection responses. Poly(lactide-co-glycilide) (PLGA) has controllable biodegradability and good biocompatibility. In this study, we evaluated 2-dimensional porcine small intestine submucosa (SIS)/PLGA hybrid films on adhesion, proliferation and phenotype maintenance of intervertebral disc cells. We prepared natural/synthetic hybrid films using 0, 10, 20, 40 and 80 wt% of SIS by the solvent evaporation method. SIS/PLGA films were characterized by scanning election microscopy (SEM). Disc cells were seeded on PLGA and SIS/PLGA film and we confirmed the effects of adhesion and proliferation on disc cells according to content of SIS by cell count and SEM observation. Contents of 10 wt% SIS positively affected to proliferate and maintain of disc phenotype in SIS/PLGA Film.

Demineralized bone particle(DBP) that affects to cell proliferation and differentiation has been used to clinical trial as natural material. hyalunonic acid(HA) called natural moisturizer can be used as biomaterials. In this study, we tried to design and characterize novel DBP-HA sponge. DBP-HA sponge were prepared by freeze-drying method after addition of 1 wt% HA solution into fabricated DBP sponge. Sponge was crosslinked with 1-ethyl-(3- 3-dimethyl aminopropyl) carbodiimide hydrochloride(EDC) solution with 50 mM concentration for 24 hrs and lyophilized. We seeded cells in DBP-HA sponge. Cellular attachment was assayed by 3-(4,5-dimethylthiazole-2-yl)- 2,5-diphenyltertazolium-bromide(MTT) test. DBP-HA sponges were characterized by scanning electron microscopy, fourier transeform infrared spectrometer and water ubsorption ability. We demonstrated presence of HA in DBP-HA sponge from C-O functional group observed by the FT-IR analysis. Moreover, we confirmed high water uptake ability and high cell viability in DBP-HA sponge. Therefore, we could expect that DBP-HA scaffolds are applicable for the tissue regeneration.

Even though the steroid drug dexamethasone is an efficacious antiinflammatory drug, it is difficult to formulate in an injectable formulation because of its poorly water solubility. So, polymeric film containing dexamatasone was designed drug delivery system including small intestinal submucosa (SIS) and sustained release of the drug from the polymeric system. poly(D,L-lactide-co-glycolide)(PLGA) is used for polymeric film that has been widely used as carriers in controlled release delivery system due to its biodegradability and relatively good biocompatibility. Also, the release behavior of dexamethasone on film controlled by adding the additive such as SIS. SIS derived from the submucosa layer of porcine intestine has been widely used as biomaterial due to low immune response and biocompatibility. SIS/PLGA film were manufactured by the method of solvent and characterized by scanning election microscopy (SEM). In vitro release behavior measured by HPLC and the PLGA molecular weight according to degradation of PLGA analyzed by GPC and pH meter. In this study, the films containing natural materials such as SIS according to content showed zero-order release profile. It was confirmed that the release of dexamethasone form films could be easily controlled by adding SIS.

The cell and porous scaffold are main components of tissue engineering. The cell seeded-porous scaffold is cultured in vitro to make tissue-like structure and then transplanted into in vivo to reconstruct the injured tissue and organ. However, in vitro culture method can not supply sufficient blood vessels and nutrients and growth factors for tissue formation and also require long cell culture time. For these reasons, in vivo culture method using omentum has been studied, because the omentum can supply many blood vessels and various nutrient and oxygen. But once the porous scaffold transplanted into in vivo, the intrusion of other kinds of cells into scaffold pores and the excessive accumulation of extracellular matrix are inevitable. As a result merits of the use of omentum are not worth. To avoid these problems and use omentum effectively, stable cell layer composed with cells have to be made. In this experiment, we made stable cell layer using PLGA sheet and used Q dot for identification of in vivo cell distribution.
